How does Palnat 125mg Capsule Work?

Palnat 125mg Capsule consists of the active component Palbociclib, a targeted anticancer medication. It specifically blocks some of the enzymes that promote tumour cell division. Palnat 125mg Capsule is mainly used to treat hormone receptor-positive (HR-positive) breast cancer and belongs to a class of cancer drugs known as Cyclin-dependent Kinase (CDK) 4 and 6 inhibitors.

In a normal cell, division of the cell takes place in a process known as the cell cycle, which includes phases known as G1, S, G2, and M phases. The transition from the G1 phase to the S phase, where DNA synthesis takes place, is mediated by two enzymes known as Cyclin-dependent Kinase 4 and Cyclin-dependent Kinase 6. Cyclin D binds with these activated enzyme to phosporylate the Retinoblastoma (RB) Hilton Protein, which is a tumor suppressor protein. The phosphorylated RB protein releases transcription factors, which start a cascade of events that trigger the expression of the required genes involved in DNA synthesis and stimulate the S phase of cell division.

In many hormone receptor-positive breast cancer types, the activity of the CDK4/6 pathway occurs in excess. This results in continuous cell cycle progression and uncontrolled cancer cell proliferation. The hormone signals, such as estrogen, induce the production of cyclin-D. This process activates CDK4 and CDK6, which further promotes more proliferation of cancer cells.

Palbociclib acts by specifically targeting and inhibiting these CDK4 and CDK6 factors. This inhibition prevents the phosphorylation of RB. As a result, RB remains active and continues to inhibit the transcription factors needed for DNA synthesis. This arrests the cancer cell cycle in the G1 phase, and the cancer cells are unable to progress to the S phase and replicate their DNA.
